Description: Recent studies have shown antimicrobial peptide (AMP) excreted by fish with its skin mucus possess anticancer activities. In this study, we are interested in characterised and evaluating the antibacterial and anticancer activity of the AMPs excreted by Anabas testudineus, since this fish is very hardy, hypothetically attributed to the secretion of its mucus. Heat de-activated Pseudomonas aeruginosa was used to challenge A. testudineus, increasing the mucus production increased more than 100% after 10 days challenge. The mucus was then extracted and subjected to antibacterial activity testing by the disc diffusion method to confirm the fish��s AMPs production. The crude mucus extracted were able to inhibit Escherichia coli, and P. aeruginosa (inhibition zone were 10 � 0.23 mm and 12 � 0.43 mm, respectively) with non-potential effect on Bacillus subtilis and Bacillus cereus and by using MTT assay showed cytotoxic effect against human breast cancer cell lines MCF7 and MDA-MB-231 (IC50 were 4.27 � 0.15 mg/ml and 4.97 � 0.25 mg/ml). Mucus was then fractionated, and cytotoxic activity of each fractioned was screened for cytotoxicity effect. Fraction 2, which shows toxicity against MCF7 and MDA-MB-231 cell lines, and antibacterial activity against (E. coli, P. aeruginosa, B. subtilis, and B. cereus) were sent for peptide sequencing to identify the bioactive peptide.
