Description: | Shiitake (Lentinus edodes) extracts have been reported to have anticancer activities however the efficacy and the pathway involve remains elusive. To determine the bioactive compound(s) within the Shiitake extracts involved in repressing malignant human melanoma (A375.S2), an array of Shiitake mutant was developed via γ-irradiation. Potential spores from each potential mutant were cross bred to obtained stable mutants. Out of 15 mutant line dikaryon mycelium (MLDM) A37 has been shown the highest growth performance stability and A37 has been used for extraction. To screen for any anti-melanin activity, zebrafish embryos have been treated with both A37 and wild mycelium (WE) extract. Both A37 and WE (200 μg/ml) significantly reduced the pigmentations in tail of the zebrafish embryo at 42.33±2.51 pigmentation spots/tail (A37) and 39.33±1.00 pigmentation spots/tail (WE), respectively compared to untreated control (63.33±2.08 pigmentation spots/ tail). Both WE and A37 have shown anti-proliferation effects towards A375.S2 (IC50 = 890 μg/ml and 749 μg/ml, respectively), which interestingly did no repress the proliferation of normal control cells, NIH3T3 (fibroblast from mouse embryos) and HS27 (Human new born foreskin fibroblast). WE show the ability to arrest the cell cycle of A375.S2 at G0/1 phase suggesting the extract targets the cell cycle checkpoint pathway. Further immunoblot studies reveal that this extract cause cell cycle arrest by inhibit the phosphorylation of Glycogen synthase 3 - beta (pGSK3-β). In conclusion, the shiitake crude extracts maybe a viable therapeutic agent to be used against melanoma, however further clinical studies need to be conducted to confirmed this findings.,Tesis ini tidak ada Perakuan Tesis Sarjana/Doktor Falsafah,Ph.D. |
