Title: Anti-trypanosomal assessment of artemisia herba-alba and osmarinus officinalis extracts following intraperitoneal infection in rabbits: clinical, parasitological and haemato-biochemical responses

Description: Artemisia herba-alba and Rosmarinus officinalis are two medicinal plants with potential for therapy of Trypanosomiasis. This study was conducted to determine the antitrypanosomal properties of crude chloroform extracts of A. herba-alba (CCEAHA) and R. officinalis (CCERO) as well as crude ethanol extracts of A. herba-alba (CEEAHA) and R. officinalis (CEERO) following intraperitoneal exposure to infected rabbits. A total of 30 New Zealand white rabbits in each set of experiment were divided into six major groups, namely pre-infection (Group 1), concurrent (Group 2), post-infection (Group 3), positive control (Group 4), negative control (Group 5) and no treatment and infection (Group 6). Animals in group 1 were given intraperitoneally (I.P) 100 mg kg-1 of extracts two days before T. evansi infection until 4 days after infection. Group 2 rabbit was infected and concurrently treated with 100 mg kg-1 of extracts for 6 days. Rabbits in Group 3 were treated with 100 mg kg-1 of extracts for 6 days after detection of parasitemia. Group 4 as positive control was treated intramuscularly (I.M) with 3.5 mg kg-1 Berenil® after establishment of parasitemia following T. evansi infection. Group 5 as negative control was not treated but infected, Group 6 served as reference group and treated by I.P. with Alsever's solution and not infected. The course of T. evansi infection in rabbits was followed for 48 days post-infection (DPI). Rabbits were closely examined for clinical observation. Blood samples were taken to measure haematological and biochemical parameters. Our results indicated that animals in group 5 showed prominent clinical signs that were characteristic of T. evansi infection with significant levels of parasitemia. Moreover, increased levels of alanine transaminase (ALT) and aspartate transaminase (AST) with mean concentration of (157.07±34.61 & 188.98±75.22) respectively were also detected in the negative control group in comparison to the reference control with mean levels of (40.57±3.24 & 99.54±16.33) respectively. In contrast, decreased levels of blood indices; notably packed cell volume (PCV), haemoglobin (Hb) and red blood cells (RBC) were recorded with mean concentration of (10.60±2.30, 74.94±7.07 & 2.46±.33) respectively in comparison to the reference group with mean concentration of (35.80±1.64, 131.80±2.16 & 4.53±.66) respectively. Animals treated with extracts of A. herba-alba and R. officinalis showed normalization in almost all investigated parameters that include clinical manifestation, haematological and biochemical alterations. Among treated groups, however, A. herba-alba showed predominantly significant antiparasitic properties in comparison to R. officinalis. Among the extracts, the chloroform extracts possessed predominantly protective activities in comparison to that of ethanol extracts. In conclusion, the manipulation of the extracts prevented severe clinical signs, haemotological and biochemical alterations, as there was relatively normalization in these parameters thereby confirming the antiparasitic properties of A. herba-alba (CCEAHA & CEEAHA) and R. officinalis (CCERO). The results data suggest that extracts of A. herba-alba and R. officinalis possess effective antitrypanosomal activity, and reinforce the use of these plants as an alternative remedy as traditional medicine against T. evansi infection.,Certification of Master's/Doctoral Thesis" is not available
