Description: The present study was designed to evaluate the effects of Rosa damascena extract on male rats fertility and determine the bioactive compounds present in the extract. Preliminary screening was performed to confirm its enhanced-fertility properties compared to Tribulus terrestris, a plant used in Iran to enhance male fertility. Based on sperm count and testosterone levels, the R. damascena hexane extract was found to exhibit the best activity (71% of control, 2.95 ± 0.12 ng/mL respectively) compared to the hexane extract of T. terrestris (31.2% of control, 1.6±0.08 ng/mL respectively). The hexane extract of R. damascena was investigated further. The treatment groups were force fed with hexane extracts of R. damascena at the doses of 5, 10, 20 and 30 mg/kg body weight respectively for 45 days. Results revealed that the hexane extracts of R. damascena increased male fertility at all dosages, with the best dosage at 20 mg/kg body weight that increased sperm count, sperm motility, sexual behavior and testosterone level in the blood of treated rats. Histologically, testes in treated groups showed very successive stages of spermatogenesis compared to control. The hexane extract was fractionated and the biological activity of all fractions was evaluated using suitable bioassays. In the most active fraction where the sperm count was (8.57±0.78) x106 compared to the control which was (5.21±0.86) x106, the major compound was identified as eugenol based on the results of GC/MS and GC/MS TOF analyses. Experiments on an authentic sample of eugenol showed that the increase in the sperm count was similar to the active fraction of R. damascena (8.86±0.95) x106. The effects of eugenol on the reproductive system of the rats are also related to its antioxidant properties. Antioxidant enzyme activity in testes, seminal vesicle and epididymis were analysed to determine the possible mechanism involved in fertility-enhancing properties. Antioxidant enzyme assays showed that the bioactive compound increased activity of catalase, glutathione peroxidase and glutathione reductase. Coadministration of the bioactive compound with nicotine (antifertility compound), showed that the bioactive compound was able to improve the fertility of nicotine treated rats. This study showed that the eugenol in R. damascena increase the fertility of male rats as evidenced by the sperm count, testosterone level, sexual behavior, antioxidant activity and testis histology data.,PhD
