Description: | Ficus carica or 'fig' is an important medicinal and economical plant which is the member of the genus Ficus. It is an important source of natural antioxidants. As the synthetic antioxidants widely used in foods have been suspected to cause detrimental effects on health, so there is a growing interest in studies employing natural additives as potential antioxidants. In solving the above-mentioned problem, the total phenolic content and the antioxidant activity of Ficus carica Linn latex from eighteen cultivars and essential oil from four samples were investigated in this study. In this investigation, the total phenolic content of latex and essential oil was calculated using Folin-Ciocalteu assay. The 1,1-Diphenyl-2-picrylhydrazyl (DPPH); 2,2'-azinobis-(3- ethylbenzothiazoline-6-sulfonic acid) (ABTS) and ferric-ion reducing antioxidant power (FRAP) assays were used for the determination of antioxidant activity. Maceration extraction process and ultrasound-assisted extraction (UAE) process were used for extraction of the antioxidant and total phenolic content from fig tree latex using 75% ethanol as solvent. The essential oil was extracted using steam distillation method. Response surface methodology was used for the optimization of antioxidant activity and total phenolic content of latex of F. carica (White Genoa cultivar). The bioactive compounds from latex were identified and quantified using liquid chromatography (LC) coupled with quadrupole-time-of-flight (QTOF)-mass spectrometer (MS/MS). Gas chromatography mass spectrometer (GC-MS/MS) was used to identify and quantify the volatile compounds from essential oil. Under optimum conditions, the latex of (White Genoa) cultivar showed the highest antioxidant activity of 65.91 ± 1.73% and 61.07 ± 1.65% DPPH; 98.96 ± 1.06% and 83.04 ± 2.16% ABTS; 27.08 ± 0.34 and 24.94 ± 0.84 mg TE/g latex in FRAP with maceration and ultrasound-assisted extraction process respectively. Total phenolic content of the White Genoa cultivar is 315.26 ± 6.14 and 298.52 ± 9.20 g GAE/mL for maceration and UAE processes respectively. On the other hand, the essential oil of the Panachee cultivar showed 93.79% and 99.31% inhibition for the DPPH and ABTS assays with 988.57 and 719.04 g TE/ml oil of trolox equivalent antioxidant capacities, respectively. Among the two extraction processes, maceration extraction shows highest performance to extract the bioactive compounds from fig latex. The overall results of this research demonstrated the potential of various cultivars of F. carica L. to be a source of natural antioxidants. This is suitable for further development in the areas of dietary supplements and various food additives. The latex and volatile oils present in F. carica L. contain bioactive compounds such as vanillic acid, phytol, and octacosane with health promotion properties, brings about the possibility of it being used for the synthesis of different drugs in the future.,Ph.D. |
