Description: | Chemo-mechanical debridement is crucial in endodontics. Sodium hypochlorite (NaOCl), despite being the gold standard endodontic irrigant, is associated with several disadvantages including a reduction in root dentine microhardness. Effective eco-produce (EEP), namely orange-pineapple mixture (M-EEP) and papaya (P-EEP), are potential endodontic irrigants due to their hydrolytic and antimicrobial properties. This research aims to determine the most effective concentration of EEP against Enterococcus faecalis and determine the effect of EEP on dentine microhardness. The minimum inhibitory concentration (MIC) of EEP, fermented for three months and six months duration respectively, was determined using broth microdilution procedure; followed by minimum bactericidal concentration (MBC) determination on brain heart infusion agar plate. EEP concentration showing MBC at three months fermentation, along with 2.5% NaOCl as the positive control, were used as irrigants on eight root halves of single-rooted human premolar teeth in each group. The Vickers Microhardness Tester with 25 g load and 10 s dwell time was used to determine the microhardness at coronal, middle and apical root regions, before and after immersion into respective irrigant for 5 minutes. All EEP showed MIC at 50% and MBC at 100% concentration. Though EEP at 50% and 100% concentrations showed higher Enterococcus faecalis inhibition compared to 2.5% NaOCl, however, no significant difference was noted between groups. Independent-samples T-test showed no significant difference in Enterococcus faecalis inhibition between M-EEP fermented for three months and six months duration but P-EEP exhibited significant Enterococcus faecalis inhibition at six months fermentation compared to three months at concentrations of 25% (p=0.017) and 0.78% (p=0.009). 100% M-EEP, 100% PEEP, and 2.5% NaOCl caused a significant reduction in dentine microhardness at all root-third regions but the greatest reduction was at the coronal root-third region compared to the middle and apical regions as shown by the two-way mixed Analysis of Variance. The magnitude of microhardness reduction was significantly lower with 100% M-EEP (4.98+2.39) compared to 100% P-EEP (7.59+2.63) and 2.5% NaOCl (7.56+2.32), p=0.002. 100% P-EEP and 2.5% NaOCl showed a similar magnitude of reduction (p=1.000).
