Description: Cell-based bone tissue regeneration is actively being investigated as an alternative treatment for paediatric bone disease. However, limited studies are available in understanding the amount of regenerated bone at different time frames which is important in the clinical management of patients. The aim of this study is to compare the bone volume regenerated using stem cells from human exfoliated deciduous teeth (SHED) with granular hydroxyapatite scaffold (Granumas®) in rat maxillary bone at 3 and 6 weeks. Pre-osteoblast cells (MC3T3-E1) were used as control in this study. Quantitative Polymerase Chain Reaction (qPCR) was employed to investigate the expression of osteoblast gene markers in control cells and mesenchymal cell markers in SHED. Both MC3T3-E1 and SHED were then seeded in Granumas® scaffold and transplanted in an artificial bone defect of 4mm in the rat maxilla. Three-dimensional (3D) analysis using micro-computed tomography (CT) and histological evaluation were performed. MC3T3-E1 cells showed positive expression of Osteocalcin (OCN) and Collagen Type 1 (Col1α1) and SHED expressed CD73, CD105 and, CD146 surface markers. Morphometric analysis showed no significant differences (p>0.05) of bone volume ratio (BV/TV) amongst the SHED and control groups after 3 weeks of transplantation. However, there were significant changes after 6 weeks (p<0.05). In histological evaluation, new bone formation was observed in all the groups. SHED showed less new bone generation compared to MC3T3-E1 group but the rate of the regenerated bone increases from 3 to 6 weeks was more. In conclusion, SHED seeded in Granumas® generated almost equal amount of bone compare with other groups. This is an advantage since SHED is easier to be obtained compared to other stem cells and present as alternative option for autologous bone graft.
