Abstract: | Pseudomonas resinovorans CA 10 is a degradative bacterium that was isolated from activated sludge. It possesses several enzymes that affect the degradation of carbazole (CAR), including extradiol dioxygenase (CarBaBb) and hydrolase (CarC). Nevertheless, it is unclear whether these enzymes are present intracellularly. We used the Biacore system to determine whether CarBaBb and Care interact and form complexes. This study assessed the affinity and kinetics of the interaction between CarBaBb and Care using Biacore. We obtained CarBaBb and Care by affinity chromatography. We then determined the affinity between CarBaBb and Care. The Biacore results indicated that CarBaBb and Care do not interact, suggesting that these enzymes do not form complexes. Therefore, such complexes do not exist within P. restinovorans cells, or any complex that does exist involves other compounds or enzymes. |
