Abstract: Platelet aggregation is important/actor which contribute in forming thrombus because of uncontroled clotting blood. Thrombus can cause the plug on coroner artery and blood vessel in brain. An anti platelet agent is a member of a class of compound that decreases platelet aggregation and inhibits thrombus formation. Many agonists of platelet aggregation such as arachidonic acid (AA), adenosine diphosphate (ADP), collagen (COL), epinephrine (EPN). The objective of this research is to determine the in vitro antiplatelet activity of four fraction respectively hexan, ethyl acetate, butanol and water fraction of velvet bean seed (M pruriens L.) used epinephrine (EPN) as agonists of platelet aggregation. The anti platelet activity were tested in human platelet rich plasma with hyperaggregation. To determine the antiplatelet activity, EPN-induced platelet aggregation was arranged at 4 level concentrations respectively 300 mM; 150 mM, 75 mM and 30 mM; the concentration of hexan, ethyl acetate, butanol, wafer fraction and aspirin as positive control were 500 Jig/mL. The results showed that wafer, butanol and ethyl acetate fraction of velvet bean seed exhibited potent inhibition of platelet aggregation induced by all concentration EPN. The strongest antiplatelet was water fraction and had the same antiplatelet activity with aspirin as positive control at level 150 mM, 75 mM and 30 mM of EPN inducer. Butanol fraction had same antiplatelet activity with aspirin at lowest EPN inducer (30 mM).
