Abstract: | Edible Bird’s Nest (EBN) is a widely known food among Asia countries due to its high nutritional content. EBN is a glycoprotein produced from the saliva secretions of some specific species birds, for example the genus Aerodramus and Collocalia. During the processing of EBN, by-products that still consist of glycoproteins are discarded which causes huge economic loss. In this study, clean EBN (SBWb) and EBN by-products (SBWps) were hydrolysed using alcalase enzyme to produce clean EBN hydrolysate (SBWhb) and EBN by-products hydrolysate (SBWhps). The purpose of this study was to determine the properties of EBN and EBN hydrolysates, the effects of the different purification methods on its properties, recovery of EBN hydrolysate fractions and to determine their physicochemical properties and antioxidant activities. EBN hydrolysates were fractionized using 2 different methods, which were cross flow filtration (FLUX) and anion exchange chromatography (AEC). SBWhb had shown to have significantly highest value (p<0.05) chemical properties, ABTS value and FRAP value whereas SBWhps had significantly (p<0.05) highest DPPH value with 62.54 % amongst the sample of EBN and EBN hydrolysate. Besides that, the first fraction of SBWhb fractionated by AEC (CF1) showed the significantly (p<0.05) highest values for soluble protein, total polysaccharide, sialic acid, reducing sugar contents and FRAP value at 22.86±2.41 mg/g, 16.70±2.06 mg/g, 101.60±12.8 mg/g, 39.64±5.67 mg/g and 17.44±1.58 ppm respectively; whereas the fifth fraction of SBWhb (CF5) had shown the highest value for OPA analysis significantly with the value of 2736.73±99.5 mg/g. From these data, CF1 and SBWhps with the highest activities were analysed by NMR to determine the structure. In conclusion, this study has successfully purified EBN hydrolysate from EBN processing by-products and the research findings showed potential application in the EBN industry by improving and diversification of the EBN products in the market. |
