Description: The effects of temperature, pH and substrate concentrations on biobutanol production by Clostridium acetobutylicum NCIMB 13357 using date fruit extract as the fermentation medium was studied. The production of biobutanol using different concentrations (from 10 gL-1 to 100gL-1) date fruit seed was also studied. Enhancement of biobutanol production by C. acetobutylicum NCIMB 13357 using different media with date fruit as the carbon source was also studied using Reinforced Clostridial Medium (RCM), Monot��s medium, and P2 medium. Further enhancement of biobutanol production by C. acetobutylicum NCIMB 13357 using P2 medium with different concentrations of vitamins such as biotin, para-aminobenzoic acid and thiamine was established. Results showed that medium containing 30 gL-1 of date fruit juice at 35oC incubation temperature with initial medium pH of 7.0 � 0.2 gave the highest concentration of ABE of 3.1, 0.1, and 1.1 gL-1 of butanol, ethanol and acetone, respectively. The yield and productivity of biobutanol were 0.32 g g-1 and 0.04 gL-1h-1 respectively, while the yield and productivity of ABE were 0.45 gg-1 and 0.06 gL-1h-1, respectively. The highest concentration of biobutanol produced (1.0 gL-1) was accomplished with 100 gL-1 of the date fruit seeds concentration. The highest productivity and yield of biobutanol were 0.02 gL-1h-1, and 0.21 gg-1, respectively when date fruit seeds concentration of 100 gL-1 was employed. The productivity and yield of ABE were 0.02 gL-1h-1, and 0.25 gg-1 respectively at the same date fruit seeds concentration. Using a combination of RCM-date fruit medium, the effects of temperature, pH and concentration of date fruit juice on the efficiency of C. acetobutylicum to produce biobutanol showed that 40 gL-1 date fruit filtrate at initial pH of 7.0 and temperature of 35 oC were the most suitable fermentation conditions that gave the highest concentration of butanol, 4.4 g L-1. The yield and productivity of biobutanol were 0.3 gg-1 and 0.07 gL-1h-1, respectively while the yield and productivity of ABE were 0.41 gg-1 and 0.09 gL-1h-1, respectively. Results when a combination of Monot��s-date fruit juice medium was used, showed that 40 gL-1 of date fruit juice at initial pH 7.0 � 0.2 and temperature of 35 oC were the best conditions for fermentation process to produce 5.31 gL-1 and 8.42 gL-1 as biobutanol and ABE and the yield and productivity of ABE was 0.52 and 0.12 gL-1h-1, respectively. When a combination of P2-date fruit juice medium was used it was found that the best date fruit juice concentration was 50 gL-1 and 11.0 gL-1 of biobutanol produced, the yield and productivity of biobutanol were 0.48 gg-1 and 0.16 gL-1h-1, respectively. The additions of vitamins to a combination of P2-date fruit juice medium significantly enhance biobutanol production. Results showed that, when P2- date fruit juice medium was supplemented with 0.02 mgL-1 of biotin, the biobutanol and ABE production reached to 14.9 gL-1 and 18.8 gL-1, respectively. In conclusion, a combination of P2-date fruit juice medium supplemented with extra concentration of biotin was the best medium to produce biobutanol using date fruit juice as the carbon source.,Certification of Master's/Doctoral thesis"is not available
