dc.description | A wide range of medicinal plant parts are used in traditional medicine practices to treat various diseases and they are also important source of potent and powerful drugs. Plants of the genus Ficus belonging to the family Moraceae are well known as source of biologically active compounds. In the present study, the methanol extracts of stems of F. aurantiaca and F. parietalis, and leaves of F. annulata showed strong immunomodulatory, xanthine oxidase and soybean lipoxygenase inhibitory activities. Phytochemical investigation on these three species of Ficus revealed the presence of three new triterpenoids, 28,28,30-trihydroxylupeol (115), 3,21,21,26-tetrahydroxy lanostanoic acid (116) and dehydroxybetulinic acid (117), one new sterol ficusterol (118) and one new carboxylic acid, 4,5-dihydroxypentacosanoic acid (119) in addition to thirteen known compounds i.e. taraxerone (32), taraxerol (31), ethyl palmitate (120), herniarin (121), stigmasterol (111), ursolic acid (29), acetyl urslic acid (27), 3,23-dihydroxylanostanoic acid (122), 3-acetylbetulinic acid (38), stigmasta-4,6-diene-3-ol (123), oleanolic acid (36), betulinic acid (26), and friedelinol (37). The new triterpenoids, 115, 116 and 117 were isolated from the stem of F. aurantiaca. The new sterol, 118 was isolated from the leaves of F. annulata while the new carboxylic acid, 119 was isolated from the stem of F. parietalis. The structures of the compounds were elucidated by various spectroscopic techniques. All the compounds were investigated for their effects on reactive oxygen species (ROS) production using luminal-based chemiluminescence assay and on isolated polymorphonuclear leukocytes (PMNs) chemotaxis using Boyden chamber technique. Xanthine oxidase and soybean lipoxygenase enzyme inhibitory activities were evaluated spectrophotometrically. Among the compounds tested, eight compounds (115, 32, 120, 121, 116, 117, 29 and 119) were very active against PMNs chemotaxis towards the chemoattractant Nformyl-methionyl-leucyl-phenylalanine (fMLP) with IC50 values of 6.8. 4.1, 3.7, 8.2,2.8, 3.6, 2.5 and 6.2 μM respectively, comparable to that of the positive control ibuprofen (6.7 μM). Seven compounds (32, 120, 121, 116, 29, 117 and 119) showed remarkable inhibition of ROS production of isolated PMNs with IC50 values of 1.3,1.1, 0.5, 0.9, 0.8, 0.9 and 5.8 μM respectively, which were lower than that of aspirin (9.4 μM). Eight compounds (115, 32, 120, 121, 116, 117, 29 and 119) exhibited strong inhibition of ROS production in human whole blood with IC50 values of 11.5, 4.6, 0.7, 1.6, 1.6, 1.3, 1.7, 3.9 μM, respectively. Sixteen compounds showed strong inhibition against soybean lipoxygenase enzyme where five compounds (32, 120, 121, 26 and 119) were very active with IC50 values of 0.9, 1.3, 1.4, 1.6, 1.2 μM, respectively. Seven compounds (32, 121, 116, 29, 117, 26 and 119) showed very strong inhibition on xanthine oxidase enzyme with IC50 values of 2.4, 6.1, 5.9, 2.6, 7.1, 6.1 and 1.5 μM respectively. These results suggest that some of the isolated compounds might be a prospective source of new leads for the development of new immunomodulatory agents to modulate the innate immune response of phagocytes and also new antiinflammatory agents.,Ph.D. | |
