dc.description | The wound healing property of Moringa oleifera leaf (MOL) extracts or locally known as 'daun kelor', has been well demonstrated experimentally in both in vitro and in vivo models. However, there is a lack of research which focuses on formulating MOL into a functional wound dressing. This research focused on the development of standardised MOL hydrocolloid dressing and followed by in-vitro and in-vivo wound healing evaluation on diabetic rat models. Firstly, the MOL was extracted in various solvents (aqueous, 50, 70 and 100 % ethanolic extracts) respectively. The extracts were screened and standardised with the reference standards using UHPLC-LCMS technique. The extracts were then tested for in vitro biocompatibility and cell migration using human dermal fibroblast (HDF) and epidermal keratinocytes (HEK) cell lines. MOL aqueous extract was then formulated into alginate-pectin (SA-PC) based hydrocolloid film and characterised for the physicochemical properties (mechanical properties, swelling ratio, moisture vapour transmission and moisture absorption studies) and the bioactives released from MOL extract-loaded hydrocolloid dressing using Franz diffusion cells. This was then followed by evaluation of in-vivo wound healing efficacy of MOL standardised aqueous extract-loaded hydrocolloid films in increasing dose strengths (0.1, 0.5 and 1%) on streptozotocin-high fat diet (STZ/HFD) induced diabetes type II rat model. Based on UHPLC results, all MOL extracts were found to contain vicenin-2, chlorogenic acid, gallic acid, quercetin, kaempferol, rosmarinic acid, and rutin. Rosmarinic acid also found for the first time in MOL extracts. Among all MOL extracts, the standardised aqueous leaf extracts exhibited the highest cell viability in HDF and HEK more than 99 % (p<0.05) compared to the untreated group after 24 h exposure as well as the migration properties (wound gap closure within 48 h) was fastest at a concentration of 12.5 μg/mL. It was suggested that this may attribute to the high amounts of vicenin-2 and chlorogenic acid in MOL aqueous extract compared to other ethanolic extracts. In the film pre-formulation, alginate-pectin (SA-PC) film was further selected to be formulated with standardised MOL aqueous extract (0.1, 0.5 and 1 %) with the highest potency in vitro migration. In acute dermal toxicity studies, all MOL film formulations exhibited no signs of dermal toxicities compared to control group. In excision wound model, 0.5 and 1 % MOL film significantly enhanced the wound closure by 77.67 ± 7.28 and 66.56 ± 12.67 % respectively at day 7 than diabetic untreated control (16 ± 3.82 %) (p<0.05). While in abrasion wound model, the commercial dressing, and 0.5 % MOL film accelerated the wound healing significantly by 73.13 ± 8.05 and 81 ± 4.5 % respectively. Based on the hydroxyproline content in wound biopsies, 0.5 % MOL film group exhibited increased collagen content by the significant higher amount (p<0.05) of hydroxyproline content (63.17 ± 8.69 μg/μL) at day 21 of wound induction period compared to diabetic untreated control group (46.85 ± 14.53 μg/μL). As a conclusion, a SA-PC film wound dressing containing standardised MOL aqueous extract was successfully developed. Based on the evaluation of in vivo diabetic wound healing, the 0.5 % MOL standardised aqueous extract-loaded hydrocolloid film is the most promising approach for treating both diabetic excision and abrasion wound.,Ijazah Doktor Falsafah | - |
