dc.descriptionPhyllanthus amarus Schum. & Thonn. is widely used in traditional medicine for the treatment of many inflammatory diseases. This study was performed to investigate the cytotoxic effect of 80% ethanol extract of P. amarus and its active compounds in HCT 116 human colon and MCF-7 human breast cancer cell lines, and the immunomodulatory effects of their generated lysates on antigen presenting cells (APCs). The apoptotic effect was quantified by Annexin V/propidium iodide co-staining using flow cytometer. Caspases 3/7, 8 and 9 activities were measured by colorimetric assay. The expression of cleaved poly ADP ribose polymerase enzyme (PARP) and cytochrome c proteins was investigated by immune-blot technique. Tumor lysate (TLY) generated by P. amarus extract was used as tumor associated antigen (TAA) to impulse the APCs including dendritic cells (DCs). The effect of the P. amarus-generated TLY was evaluated on the antigen presentation capacity of APCs, mainly the expression of MHC class I, II as well as the co-stimulatory molecules CD 83 and 86. P. amarus was analyzed by using a validated reversed-phased HPLC and LC-MS/MS. P. amarus induced apoptosis in both cell lines and the caspases activities increased concentration-dependently. It upregulated the expression of cleaved PARP (Asp 214) and cytochrome c. Among the compounds, phyllanthin and gallic acid exhibited significant cytotoxic activities against both cell lines. DCs exhibited the highest antigen presentation capacity among the APCs and markedly upregulated the expression of co-stimulatory molecules CD 86 and CD 83. The release of IL-12 and IL-6 were enhanced by TLY-treated DC at ratio of 1 DC: 3 tumor cells, however, the release of anti-inflammatory cytokine IL-10 was suppressed. The phagocytic capacity of TLY-pulsed DCs was suppressed and the chemotaxis capacity and T-cell proliferation activities of APCs were significantly enhanced. The gene expression profiles for cytokines of the APCs showed increase in the mRNA gene expression of IL-12 and IL-6 in tumor lysate-DCs. In conclusion, P. amarus extract and its active compounds demonstrated cytotoxic effects and the influence of P. amarus-generated TLY on immune effector mechanisms of DCs showed its potential to induce an anti-tumor immune response against the recognized tumor antigen.,Doktor Falsafah
