dc.description | Tumbuhan menghasilkan pelbagai metabolit sebagai tindak balas terhadap persekitaran untuk menghadapi tekanan biotik dan abiotik. Tekanan ini akan mendorong pembentukan radikal bebas yang membawa kepada kemusnahan oksidatif dalam sel. Maka penghasilan metabolit adalah untuk melindungi tumbuhan dari kemusnahan tersebut. Objektif kajian ini adalah untuk memprofil kandungan asid amino dan flavonoid di dalam Polygonum minus dari altitud dan suhu rawatan yang berbeza untuk mengkaji hubungan antara metabolit dan persekitaran. Daun P. minus dari populasi berbeza iaitu tanah tinggi dan tanah rendah digunakan dalam kajian ini. Eksperimen rawatan suhu juga dijalankan untuk mengkaji variasi kandungan asid amino dan flavonoid terhadap perubahan suhu. Penyaringan pertama dilakukan dengan menggunakan Spektroskopi Inframerah Transformasian Fourier (FTIR) untuk melihat perbezaan dari segi kumpulan berfungsi yang hadir. Kajian ke atas asid amino dijalankan dengan menggunakan Kromatografi Cecair Berprestasi Tinggi (HPLC) manakala kajian ke atas flavonoid dilakukan dengan menggunakan Kromatografi Cecair Spektrometri Jisim yang digandingkan dengan masa penerbangan (LC-TOF-MS). Analisis Diskriminan-Separa Kuasadua Terkecil (PLS-DA) dijalankan untuk melihat variasi antara populasi dan rawatan suhu. Prolina merupakan asid amino dominan yang terhasil dalam P. minus untuk kedua-dua sampel dari populasi liar dan eksperimen rawatan suhu. Manakala dalam kajian flavonoid, kelas flavonol dan flavon merupakan flavonoid yang paling banyak dijumpai. Umumnya, populasi tanah tinggi mengandungi kandungan asid amino dan flavonoid berbanding populasi tanah rendah, manakala dalam kajian rawatan suhu, rawatan suhu tinggi menghasilkan peratusan metabolit yang lebih tinggi berbanding rawatan suhu rendah. Hasil yang diperolehi menunjukkan bahawa terdapat variasi dalam penghasilan metabolit antara populasi dan pada suhu yang berbeza, seterusnya menyokong bahawa tumbuhan bertindak balas dengan menghasilkan metabolit yang berbeza mengikut keadaan persekitarannya.,Plants produce different groups of metabolites in response to their growing environment to cope with a variety of biotic and abiotic stresses. The stress will accumulate free radical that lead to the oxidative damage, thus the production of metabolites is to protect plants from this damage. The objective of this study is to profile the content of amino acids and flavonoids in Polygonum minus from different altitudes and different temperature for investigation on the relationship between plant metabolites and the environmental effects. P. minus leaves from highland and lowland were analysed in this study. Plants were also grown under controlled environment with three different temperature treatments, to study the variation of amino acid and flavonoid towards temperature effect. The initial screening was done by Fourier Transform InfraRed (FTIR) to fingerprint the variation in functional group of compounds. Different amino acids were identified using High Performance Liquid Chromatography (HPLC) while flavonoid compounds were identified using Liquid Chromatography-Time of Flight-Mass Spectrometry (LC-TOF-MS). Partial Least Squares Discriminant Analysis (PLS-DA) was performed to study the variation between the populations. Proline was the most abundant amino acids produced in P. minus in both samples from the wild populations and temperature treatment experiment. While for flavonoid analysis, flavonols and flavones were the most abundant flavonoids in P. minus in both samples. In general, highland populations contain higher percentage of amino acids and flavonoids compared with lowland populations. While for temperature treatment experiment, plants under higher temperature treatment produced more metabolites compared with lower temperature treatment. These findings show that there is a significant variation in the metabolite production between the different populations subjected to different temperatures. This reflects the metabolite changes of plants from different altitudes in response to different temperatures.,Bachelor | |
