dc.descriptionData ketoksikan sesuatu tumbuhan perubatan tropika amat diperlukan bagi memastikan produk kesihatan yang dihasilkan selamat digunakan. Kajian ini bertujuan untuk menilai tahap ketoksikan ekstrak akueus piawai Mitragyna speciosa pada tikus Sprague-Dawley serta menentukan kesan mutageniknya ke atas bakteria Salmonella typhimurium TA98 dan TA100. Ujian toksisiti akut dijalankan mengikut 2 prosedur OECD 420 dan OECD 425 yang melibatkan 7 kumpulan tikus. Tiada simptom ketoksikan yang signifikan ditunjukkan kecuali terdapat simptom keletihan dan tidur. Nilai LD50 oral akut tidak dapat ditentukan kerana tiada kematian tikus berlaku. Parameter hematologi tidak berbeza secara signifikan kecuali min kepekatan hemoglobin korpusel bagi tikus betina kumpulan 500 dan 2000 mg/kg yang berbeza secara signifikan (p<0.05). Parameter ujian biokimia juga tidak berbeza secara signifikan kecuali bagi nilai purata albumin, kalsium dan kolesterol tikus jantan kumpulan 175 mg/kg yang berbeza secara signifikan (p<0.05). Ujian toksisiti subakut oral dijalankan mengikut prosedur OECD 407 yang melibatkan 4 kumpulan tikus. Purata pengambilan makanan dan isipadu minuman tidak berbeza secara signifikan kecuali bagi tikus betina dan tikus jantan kumpulan 2000 mg/kg yang berbeza secara signifikan (p<0.05). Purata berat badan juga tidak berbeza secara signifikan kecuali bagi tikus jantan kumpulan 175, 500 dan 2000 mg/kg yang berbeza secara signifikan (p<0.05). Purata berat organ tikus tidak berbeza secara signifikan kecuali purata berat paru-paru relatif tikus betina kumpulan 175 mg/kg yang berbeza secara signifikan (p<0.05). Berat hepar mutlak dan relatif dan juga berat mutlak ginjal kanan tikus jantan kumpulan 2000 mg/kg juga berbeza secara signifikan (p<0.05). Tiada simptom ketoksikan yang signifikan ditunjukkan kecuali terdapat simptom keletihan, bulu badan gugur, diarea, salivasi air liur, tidur dan luka belakang badan. Parameter hematologi tikus kajian tidak berbeza secara signifikan kecuali bagi nilai purata variasi isipadu sel darah merah tikus jantan kumpulan 2000 mg/kg yang berbeza secara signifikan (p<0.05). Parameter biokimia tikus kajian tidak berbeza secara signifikan kecuali nilai purata kreatin kinase tikus betina kumpulan 175 mg/kg yang berbeza secara signifikan (p<0.05). Purata alanin aminotransferase, kolesterol, amilase dan urea tikus jantan kumpulan 175 dan kumpulan 500 mg/kg juga berbeza secara signifikan (p<0.05). Parameter yang lain bagi kajian toksisiti in vivo tidak berbeza secara signifikan. Ujian Ames telah mendapati bahawa 5 kepekatan dos ekstrak tidak menunjukkan kesan mutagenik ke atas kedua-dua strain bakteria. Kesimpulannya,ekstrak akues piawai M. speciosa tidak menunjukkan sebarang kematian dan kesan ketoksikan yang signifikan dan tidak bersifat mutagenik.,Toxicological data of medicinal tropical plants is much needed to ensure their health products are safe to be used. Therefore, the objectives of this study were to evaluate the toxicity level of standardized Mitragyna speciosa aqueous extract on Sprague-Dawley rats and also to determine its mutagenic effect on bacteria Salmonella typhimurium TA98 and TA100. Acute toxicity study was done based on OECD 420 and OECD 425 procedures which involved 7 groups. No other toxicity symptoms were detected except fatigue and sleep. LD50 value for acute oral cannot be determined because no death of rats occurred. Hematological parameter was not significantly different except the corpuscular hemoglobin concentration at 500 and 2000 mg/kg female rats which was significantly different (p<0.05). Biochemical parameter was also not significantly different except the albumin, calcium and cholesterol levels at 175 mg/kg male rats which were significantly different (p<0.05). Sub-acute toxicity study was done based on OECD 407 procedure which is involved 4 groups. The average food consumption and water volume were not significantly different except for 2000 mg/kg female and male rats group which were significantly different (p<0.05). The average bodyweight was not significantly different except for 175, 500 and 2000 mg/kg male rats which was significantly different (p<0.05). The weight of organ was not significantly different except the relative weight of lungs for 175 mg/kg female rats which was significantly different (p<0.05). The weight and relative weight of liver and also the weight of right kidney for 2000 mg/kg male rats was also significantly different (p<0.05). No other toxicity symptoms were detected except fatigue, body fur drop down, diarrhea, salivation, sleep and back wound. Hematological parameter was not significantly different except the average value of red blood cells volume variation for 2000 mg/kg male rats which was significantly different (p<0.05). Biochemical parameter was also not significantly different except the average value of creatine kinase for 175 mg/kg female rats which was significantly different (p<0.05). The average value of alanine aminotransferase, cholesterol, amylase and urea for 175 and 500 mg/kg male rats were also significantly different (p<0.05). Other parameters for in vivo toxicity study do not show any significant difference. The Ames test results showed that the extract had no mutagenic effects on both strain of bacteria. In conclusion, standardized M. speciosa aqueous extract did not demonstrate any deaths and major toxic effects and also not mutagenic.,Master
