dc.description | Gout is the world's most frequent inflammatory arthritis. Available gout treatments possess undesirable side effects such as cardiovascular diseases and kidney failure. Zingiber zerumbet is a famous medicinal plant and its rhizomes have been used to treat for inflammation, chest pain and rheumatic pains. The effects of Z. zerumbet rhizome on gout is not well explored. The present study was designed to investigate the possible anti-hyperuricemia and anti-gout protective effects of Z. zerumbet extract in rat model with acute hyperuricemia and gouty arthritis. In this study 60 male Sprague-Dawely rats were divided into two categories; hyperuricemia and gout respectively. Each category was divided into five groups. Rats of all groups (except vehicle and positive control group) received Z. zerumbet crude extract at 200 mg/kg and 400 mgkg doses daily for 14 days. Before the last daily extract treatment (on day 14), animals were administered with potassium oxonate (PO) at 250 mg/kg for induction of hyperuricemia, while rats of gout groups, were injected with monosodium urate (MSU) crystals solution into the ankle joint on day 11. At the end on day 14, all the rats were sacrificed and blood and tissue samples were obtained for biochemical and histological analysis. In this study, the results showed that the serum uric acid and xanthine oxidase level were significantly decreased by Z. zerumbet extract at 400 mg/kg and 200 mg/kg doses (p<0.05) compared to the hyperuricemia model group. The two doses of Z. zerumbet also significantly increased superoxide dismutase activity and attenuated 8-isoprostane production (p<0.05) compared to model group. The ankle joint diameter was significantly reduced by Z. zerumbet at 200 mgkg and 400 mgkg dose (p<0.05) compared to model group. The two doses of the extract prevented inflammation significantly (p<0.05) compared to the model group as shown in the results of the inflammatory tests; WBC count, C-reactive protein and erythrocyte sedimentation rate test. In conclusion, both doses of Z. zerumbet extract are able to protect against hyperuricemia and gout via its anti-inflammatory and anti-oxidant activities.,Master Of Health Science (Biomedical Science) | |
