dc.description | Use of endodontic sealer with antimicrobial properties is important to eliminate the residual bacteria in the root canal after endodontic treatment. However, information on antibacterial efficacy of endodontic sealers especially newly developed sealers is lacking. This research aimed to determine and compare the antibacterial and antibiofilm activity of AH Plus, iRoot SP and EndoSeal MTA against Enterococcus faecalis, Streptococcus mutans, Staphylococcus aureus, and Actinomyces viscosus. The antibacterial efficacy of AH Plus, iRoot SP, and Endoseal MTA was assessed using a modified direct contact test. The freshly prepared sealers and the set sealers at 1, 3, 7, and 14 days were exposed to Enterococcus faecalis, Streptococcus mutans, Staphylococcus aureus, and Actinomyces viscosus for an hour. Subsequently, the bacteria count was determined after plating on respective agar for 24 hours. Meanwhile, the antibiofilm activity of all the tested sealers was performed by direct contact of the endodontic sealers with the biofilm. Biofilm was prepared by collecting the subgingival plaque from a healthy adult and inoculating its suspension to 8-well cell culture slide for incubation under anaerobic conditions at 37 for 2 days, 1 week, 2 weeks, 3 weeks, and 4 weeks. Next, 0.5 mL of tested endodontic sealers were placed directly onto the biofilm for 1 hour, 1 day, 3 days and 7 days. The number of dead and live bacteria was determined by using confocal laser scanning microscopy (CLSM). All the collected data were analysed by one-way analysis of variance of freshly prepared AH Plus and EndoSeal MTA against all investigated bacterial species was significantly higher than iRootSP (p<0.001). Only EndoSeal MTA maintained its antibacterial efficacy after 24 hours of setting with significantly higher antibacterial effect against S. mutans and S. aureus at day 1 and 3 as well as A. viscosus at day 1, 3 ,7 and 14 (p<0.001). Meanwhile, all three endodontic sealers showed a significant antibiofilm against both young and mature biofilms (p<0.001). Only EndoSeal MTA exhibited stronger antibiofilm activity against 2-day-old biofilm (p<0.001). In conclusion, EndoSeal MTA exhibits stronger antibacterial efficacy iRootSP.
