dc.descriptionOrthodontic tooth movement is a complex process involving tooth and periodontium tissue which release specific enzymes and biomarkers. Aim of this study was to investigate the profile of salivary fluid enzyme activities during orthodontic treatment, i.e Lactate Dehydrogenase (LDH) for tissue inflammation, Aspartate Aminotransferase (AST) for cell necrosis, Alkaline Phosphatase (ALP) for bone formation and Tartrate Resistance Acid Phosphatase (TRAP) for bone resorption. Nineteen healthy subject (mean age of 21.5 years) with class II division 1 malocclusion who required extraction of maxillary first premolars were recruited from postgraduate orthodontic clinic. Saliva samples were collected from 19 patients that underwent canine traction using fixed appliances. Enzymes activities were measured before placement of fixed appliances (basal activity) followed immediately before and weekly canine retraction for five weeks. The specific salivary LDH, AST, TRAP and ALP activities were analysed using spectrophotometer (405nm and 340nm). No statistical significant (p>0.05) difference was noted in LDH activity between basal activity and during canine retraction period. AST showed higher activity compared to basal activity from week 0 to week 1 with statistically significant increased (p<0.05) found in week 1. ALP showed significantly higher enzyme activity compared to basal activity from week 1 to 5 (p<0.05), with the peak level at week 5. While TRAP showed significant increased enzyme activity compared to basal activity at week 2 after canine retraction (p<0.05). The profiles of enzymes activities showed that cell necrosis occurred during early phase of treatment followed by bone resorption and bone formation with less inflammation of periodontal tissue during orthodontic treatment using self ligation bracket. Thus orthodontic tooth movement can be monitored through the expression of LDH, AST, TRAP and ALP activity in saliva as biomarkers in monitoring tooth movement in self ligating bracket system.,Certification of Master's/Doctoral Thesis" is not available
