dc.descriptionJerami padi kaya dengan bahan organik, namun ianya belum banyak diterokai untuk meningkatkan kesuburan tanah pertanian. Kajian ini dilakukan untuk memencil dan mengenalpasti satu konsortium mikrob selulolitik indigenus dan peranannya sebagai pereput untuk meningkatkan kesuburan tanah, pertumbuhan pokok dan pengeluaran hasil padi. Sejumlah 81 pencilan bakteria selulolitik, sekitar kawasan sawah padi telah berjaya dipencilkan termasuklah daripada tanah (42), rizosfera (32) dan jerami padi (7). Selepas beberapa siri saringan, sebanyak lapan pencilan bakteria sellulolitik iaitu TML2R, TML8T, TML16T, TML54R, TML55R, TML69R, TML75J, TML80J dikenal pasti berupaya menghidrolisis selulosa dan xilan melalui pembentukan zon terang di sekitar koloni bakteria, apabila ditindakbalaskan dengan iodin. Melalui kaedah penjujukan 16S rDNA, pencilan tersebut dikenal pasti sebagai Bacillus cereus untuk TML 2R, TML 16T dan TML 80J, Bacillus sp. untuk TML 8T dan TML 55R, Alcaligenes faecalis untuk TML 54R dan TML 75J dan Stenotrophomonas sp. untuk TML 69R. Kesemua bakteria ini dibangunkan sebagai Cellulolytic Microbial Consortia baru-Universiti Kebangsaan Malaysia (CMC baru-UKM). Kesan saiz inokulum, tempoh eraman dan pensterilan tanah dikaji melalui pengukuran aktiviti tiga enzim penguraian; CMCase, β-glucosidase, xilanase; serta peratusan degradasi dan nisbah C:N jerami padi. Saiz inokulum 108 cfu/ml, selama enam minggu pada tanah tidak steril didapati paling berkesan dalam pereputan jerami padi. Biojisim jerami padi menurun 69.2% (w/v) berbanding kawalan dan sekata dengan penurunan nisbah C:N daripada 38.2 kepada 29.1. Nilai indeks percambahan benih padi yang dirawat CMC-UKM meningkat kepada 116% menunjukkan ianya tidak toksik malah berpotensi berfungsi sebagai agen fitonutrien dan fitoransangan. Dengan menggunakan teknik litterbag pereputan jerami padi dalam tanah didapati menurunkan nisbah C:N dari 37.5 kepada 16.4 dan biojisim jerami padi menurun secara signifikan 23.4% (w/v) dalam tempoh eram enam minggu berbanding kawalan. Biji benih padi yang disemai dalam tanah yang telah dirawat dengan jerami padi dan CMC-UKM selama enam minggu menghasilkan perbezaan bererti pada aras berat kering akar berbanding kawalan. Jerami padi yang disembur dengan CMC-UKM di plot sawah padi Tanjung Karang, dan dibiarkan selama enam minggu memberikan kesan bererti pada berat biji padi per pokok (52.8%), 1000-butir berat padi (10.9%) dan biomes (48.50%) berbanding kawalan. Ini menunjukkan bahawa CMC-UKM berpotensi tinggi dalam meningkatkan hasil padi.,Rice straw is rich in organic material but has not been utilized to improve the fertility of paddy field soils. In this study, isolation and identification of an indigenous cellulolytic microbial consortium from soil, rhizosphere and rice straw and its role in the decomposition of rice straw and in improving soil fertility, plant growth and yield of paddy were conducted. A total of 81 cellulolytic bacteria isolates were successfully isolated from surrounding paddy field area which include soil (42), rhizosphere (32) and rice straw (7). After a series of screening, eight cellulolytic bacterial isolates namely TML8T, TML16T, TML2R, TML54R, TML55R, TML69R, TML75J, TML80J were found to be able to hydrolyze cellulose and xylan as shown by a clear zone when treated with iodine. Based on 16S rDNA partial sequence analysis, these eight isolates were identified as Bacillus cereus for TML 2R, TML 16T and TML 80J, Bacillus sp. for TML 8T and TML 55R, Alcaligenes faecalis for TML 54R and TML 75J and Stenotrophomonas sp. for TML 69R. All bacteria were then developed as a consortium, known as Cellulolytic Microbial Consortium-Universiti Kebangsaan Malaysia (CMC-UKM). The effect of inoculum size, incubation period and soil sterilization were studied by measuring the cellulolytic enzyme activities of CMCase, β-glucosidase and xylanase as well as biomass degradation and C:N ratio of the rice straw. An inoculum size of 108 cfu/ml for six weeks in non-sterile soil was found to be most effective in decomposing the rice straw; the biomass decreased 69.2% (w/v) compared to control and in conjuction with C:N ratios reduction from 38.2 to 29.1. Germination index value of seeds treated with CMC-UKM was found to be 116% indicating that it is not toxic, and in fact behaves as a phytonutrient and phytostimulant. By using the litter bag technique, rice straw decomposition in soil was found to reduce the ratio of C:N from 37.5 to 16.4 and significantly decreased rice straw biomass by 23.4% (w/v) within six weeks of incubation compared to the control. Seeds sown in soil treated with rice straw and CMC-UKM for six weeks produced significant difference in the level of root dry weight, compared to control. Rice straw sprayed with CMC-UKM at Tanjung Karang paddy plots, and left for six weeks showed a significant difference in fertile grain per paddy plant (52.8%), 1000-grain weight of paddy (10.9%) and biomass (48.5%) compared to the control. This indicates that the CMC-UKM has potential in increasing rice yields.,Ph.D.
