dc.description | Lycopodiaceae merupakan tumbuhan mirip paku pakis yang primitif bagi tumbuhan tisu bervaskular yang lain. Kajian evolusi dan pengelasan famili ini masih kabur. Pengelasan terdahulu yang menggunakan data morfologi mengelaskan famili ini dalam satu genus, Lycopodium. Terkini kesemua spesies dalam famili ini dikelaskan semula dalam tiga genus: Lycopodium, Lycopodiella dan Huperzia. Kajian ini dilakukan untuk menjana maklumat tambahan bagi tujuan pengelasan dengan menggunakan jujukan DNA. Sebanyak 17 takson yang dikelaskan dalam tiga genus tersebut telah digunakan iaitu: Huperzia carinata, H. dalhousiana, H. nummulariifolia, H. hippuris, H. phlegmaria, H. phlegmarioides, H. phyllanta, H. serrata, H. aff. squarrosa, H. prolifera, H. squarrosa, H. tetrasticha, Lycopodiella cernua, Lycopodium casuarinoides, L. clavatum, L. platyrhizoma dan L. volubile. Dua spesies telah digunakan sebagai kumpulan luaran dalam kajian ini iaitu Psilotum complanatum dan P. nudum. Sebanyak lima kawasan DNA telah digunakan untuk melihat hubungan filogenetik famili ini iaitu empat daripada genom kloroplas (cpDNA) yang terdiri daripada kawasan penjarak trnL-F, intron trnL, subunit protein ribosom 4 (rps4), dan ITS kloroplas serta satu kawasan daripada genom nukleus (nDNA) iaitu kawasan ITS1. Data jujukan telah dianalisis menggunakan perisian PAUP melalui tiga kaedah iaitu Neighbour Joining (NJ), Maximum Parsimony (MP) dan Maximum Likelihood (ML). Secara keseluruhan, pohon filogeni bagi empat kawasan DNA terhadap Lycopodiaceae menunjukkan pemisahan yang jelas iaitu kumpulan A (Huperzia), kumpulan B (Lycopodium) dan kumpulan C (Lycopodiella). Begitu juga topologi bagi kawasan penjarak trnL-F juga menunjukkan pemisahan antara kumpulan A (Huperzia) dan kumpulan B (Lycopodium). Dalam kumpulan A, terdapat dua subklad utama dengan H. serrata sebagai takson dasar diikuti oleh H. dalhousiana yang mencapah terlebih dahulu. Subklad pertama mengelompokkan H. carinata, H. nummulariifolia, H. phlegmaria, H. phlegmarioides dan H. phyllanta. Subklad kedua menunjukkan H. prolifera, H. aff. squarrosa, H. hippuris, H. squarrosa dan H. tetrasticha mengelompok bersama. Kumpulan B menunjukkan L. casuarinoides dan L. volubile membentuk klad bersama manakala L. platyrhizoma telah mencapah terlebih dahulu sebelum L. clavatum. Kedudukan La. cernua dalam topologi adalah berbeza mengikut kawasan jujukan DNA. Bagi topologi rps4 dan ITS1, La. cernua merupakan takson beradik bagi Lycopodium dan merupakan takson dasar bagi intron trnL dan ITS kloroplas. Kajian menggunakan data molekul yang telah dilakukan berjaya membezakan famili Lycopodiaceae kepada tiga genus yang berbeza dan menyokong pengelasan yang dilakukan menggunakan data morfologi.,Lycopodiaceae, a fern-allies, is the primitive of all vascular plants. Its evolution and classification is still poorly understood. The old classification using morphological data, grouped all taxa in one genus, Lycopodium. However, in current classification all species are divided into three genera: Lycopodium, Lycopodiella and Huperzia. This study was done to generate additional information for classification by using DNA sequence. A total of 17 taxa classified into three genera were used in this study which are: Huperzia carinata, H. dalhousiana, H. nummulariifolia, H. hippuris, H. phlegmaria, H. phlegmarioides, H. phyllanta, H. serrata, H. aff. squarrosa, H. prolifera, H. squarrosa, H. tetrasticha, Lycopodiella cernua, Lycopodium casuarinoides, L. clavatum, L. platyrhizoma and L. volubile. Two species used as outgroups were Psilotum complanatum and P. nudum. Five regions were used to observe the relationship of this family. Four of them located in chloroplast genome (cpDNA) which are intergenic spacer trnL-F, trnL intron, ribosomal protein subunit 4 (rps4) and ITS chloroplast while one located in nuclear genome (nDNA) is ITS1. The sequencing data were analyzed by using PAUP and conducted by Neighbour Joining (NJ), Maximum Parsimony (MP) and Maximum Likelihood (ML). Overall, phylogeny trees of four DNA regions shows clear separation of group A (Huperzia), group B (Lycopodium) and group C (Lycopodiella). As well as, topologies of intergenic spacer trnL-F shown segregation in group A (Huperzia) and group B (Lycopodium). In group A, there are two main subclade with H. serrata as a basal taxa followed by H. dalhousiana diverging first. First subclade clustered H. carinata, H. nummulariifolia, H. phlegmaria, H. phlegmarioides and H. phyllanta while second subclade shows H. prolifera, H. aff. squarrosa, H. hippuris, H. squarrosa and H. tetrasticha are joined together. Group B shows L. casuarinoides and L. volubile form a clade together while L. platyrhizoma diverge before L. clavatum. Position of La. cernua in topologies are different according to the DNA regions. For rps4 and ITS1 topology, La. cernua is sister taxa to Lycopodium and basal taxa for ITS chloroplast and trnL intron topologies. Studies using molecular data had been successfully differentiate Lycopodiaceae to three different genera and supports classification by using morphological data.,Master/Sarjana | |
