dc.description | Apicomplexa is a protozoan phylum containing more than 6,000 species of mostly parasites, infecting a wide range of animals from invertebrates to mammals. This study was carried out to investigate the presence of intracellular Apicomplexa-like parasites in the organs and tissues of commercially important mollusks within the Johor Straits, Malaysia. Among the mollusks studied were gastropods Laevistrombuscanarium (Linnaeus, 1758), Strombusurceus(Linnaeus, 1758), Cymbiolanobilis(Lightfoot, 1786), Melomelo(Lightfoot, 1786), Nassariuslivescens and Naticatigrinaand the bivalves Pernaviridis(Linnaeus, 1758), Meretrixmeretrix (Linnaeus, 1758), Gafrariumdivaricatum(Gmelin, 1791) and Clausinellachlorotica (Philippi, 1849). The mollusks were randomly sampled from Merambong Shoal seagrass beds and brought back to the laboratory for histological and electron microscopy (TEM) analyses. For histological analysis, samples were processed by standard histological technique and thin sections (5 µm) of various organs were stained using a trichrome staining (hematoxylin, alcian blue and acid fuchsine) and examined under a light microscope. For TEM analyses, sampled were fixed in glutaraldehyde solution, embedded in resin and thin sections (1 µm) were stained using Toulidine blue. Studies on the gastropods found that the conch L. canarium and S. urceus showed higher infestation rate, followed by the volutes C. nobilis and M. melo. Generally the digestive gland, digestive gland duct and renal organ of the gastropods were the most infested, and no parasite was observed in the general tissues. Based on the histological analysis, it was suggested that the apicomplexa-like parasites might complete their life cycles within the digestive gland of L. canarium and S. urceus. All three stages of the Apicomplexa life stages i.e. sporocysts, gamont and trophozoites were found in the organ. Studies on the bivalves that G. divaricatum and M. meretrix showed highest infestation rate, followed by P. viridis and C. chlorotica. Results also revealed that the bivalves' gonads and digestive glands were the most infected and no infestation was observed in the mantle tissues. Furthermore, different types of Apicomplexa parasite were recorded in bivalves where P. viridis was infested by coccidian types whilst G. divaricatum, M. meretrix and C. chlorotica were infested by gregarine types of Apicomplexa. In general, different Apicomplexamorphotypes such as gamont, sporozoite and trophozoite were observed in both the gastropods and bivalves. The trophozoites were characterized by their bottle shaped and apical structure complex, the sporocysts by their thick walls, while the gamonts by their thin walls. Apart from that, the spore, early or mature oocyst and macrogamont stages of parasite were observed in the bivalves. Results from this study are very important as it provides significant baseline information on the health profile of important mollusks species. Nevertheless, further investigations are urgently needed to confirm the identification of the parasite species and their effect on the commercial mollusk population in the study area.,Master/Sarjana | - |
