Description: Acetaminophen is an analgesic that has side effects when it is taken more than therapeutic doses. One of its side effects is acute liver failure. Some plant extract have been shown to have protective antioxidant activity that leads to hepatoprotective effect. In the present study, the effects of methanolic extracts of Plantago major L. on acetaminophen-induced hepatotoxicity in male Sprague-Dawley rats were elaborated by assessing the morphological, biochemical indices and extent of oxidative stress by measuring the levels of glutathione, superoxide dismutase (SOD) and malondialdehyde (MDA). Liver injury and glycogen accumulation were observed by using hematoxylin and eosin and Periodic Acid Schiff staining. Forty-four male rats were randomly divided into 6 groups. The baseline group (BSL) was sacrificed on day 0. The rats in DMSO group received 5% DMSO on day 0 whereas hepatotoxicity was induced with a single dose of oral acetaminophen (1000 mg/kg) in APAP, PM 1000, PM 1500 and NAC groups. Groups PM 1000 and PM 1500 then received the methanol leaf extract of Plantago major orally at 1000 mg/kg and 1500 mg/kg body weight respectively for six days and Group NAC was given 900 mg/kg of N-acetyl-cysteine. The histopathological findings showed obvious difference such as cellular necrosis and inflammatory cells in groups PM 1000 and PM 1500 compared to the APAP group.The PM 1000 and PM 1500 groups which were treated with two doses of plant extract revealed significant increase in the level of SOD and glutathione (p<0.05). However, there was a significant reduction of MDA level in groups PM 1000 and PM 1500 compared to APAP group (p<0.05). In conclusion, Plantago major L. extract prevent hepatic injuries associated with acetaminophen-induced hepatotoxicity by increasing the level of antioxidant enzymes and inhibiting the lipid peroxidation.,Certification of Master's/Doctoral Thesis" is not available
