Description: Sindrom metabolik yang juga dikenali sebagai sindrom 'X' merupakan gabungan penyakit yang terdiri daripada obesiti sentral,dislipidemia, hipertensi dan hiperglisemia. Statistik oleh WHO menunjukkan bahawa suku daripada populasi dewasa di dunia ini mengalami sindrom metabolik. Kajian terhadap sindrom metabolik semakin menarik perhatian ramai para penyelidik terutama di dalam bidang farmaseutikal. Risiko penyakit hati berlemak tidak beralkoholik (NAFLD) dan steatosis pankreas semakin meningkat apabila seseorang menghidap sindrom metabolik. Sindrom metabolik berlaku akibat ketidakseimbangan pengambilan diet dan kalori. Fruktos yang merupakan bahan pemanis di dalam makanan dan minuman menyebabkan peningkatan pengambilan kalori yang seterusnya boleh menyebabkan pembentukan sindrom metabolik. Enzim 11β-hidroksisteroid dehidrogenase jenis 1 (11β-HSD1) yang memangkinkan pertukaran hormon 11-dehidrokortikosteron kepada kortikosteron pada tikus memainkan peranan di dalam mekanisme pembentukan sindrom metabolik. Kajian ini dilakukan bagi membentuk model tikus sindrom metabolik yang dirangsang oleh air minuman fruktos pada peratus kepekatan yang berbeza iaitu 20% dan 25%. Aktiviti dan ekspresi enzim 11β-HSD1 serta komplikasi sindrom metabolik seperti NAFLD dan steatosis pankreas juga dikaji. Air minuman fruktos 20% dan 25% diberikan setiap hari selama 8 minggu secara ad libitum kepada tikus Wistar jantan di samping diet tikus yang normal. Hasil kajian ini menunjukkan bahawa pengambilan air minuman fruktos 20% dan 25% yang diambil selama 8 minggu menyebabkan pembentukan sindrom metabolik pada tikus Wistar jantan serta peningkatan aktiviti dan ekspresi enzim 11β-HSD1. Komplikasi NAFLD dan steatosis pankreas dapat dilihat pada kedua-dua kumpulan ini. Kesimpulannya, kepekatan fruktos yang lebih rendah iaitu air minuman fruktos 20% yang memenuhi kriteria sindrom metabolik serta menyebabkan peningkatan yang signifikan bagi jumlah pengambilan air minuman harian, jumlah pengambilan kalori, peratus kenaikan berat badan, indeks jisim tubuh, ukurlilit abdomen, deposisi dan berat tisu adipos abdomen berbanding pengambilan air minuman fruktos 25% diambil bagi membentuk model sindrom metabolik dan komplikasi NAFLD dan steatosis pankreas pada tikus Wistar jantan melalui mekanisme peningkatan aktiviti dan ekspresi enzim 11β-HSD1.,Metabolic syndrome also known as syndrome 'X' is a combination of diseases consisting of central obesity, dyslipidemia, hypertension and hyperglycemia. Statistics by WHO showed that a quarter of the world's adult population is suffering from the metabolic syndrome. Studies of the metabolic syndrome are increasingly attracting the attention of many researchers especially in the pharmaceutical field. The risk of nonalcoholic fatty liver disease (NAFLD) and pancreatic steatosis increases as a person is diagnosed with metabolic syndrome. Metabolic syndrome is due to an imbalance of diet and calorie intake. Fructose which is a sweetener in foods and drinks increases calorie intake which in turn can lead to the development of metabolic syndrome. The enzyme 11β-hydroxysteroid dehydrogenase type 1 (11β-HSD1) which catalyzes the conversion of 11-dehydrocorticosterone hormone to corticosterone in rodents plays a role in the mechanism of the development of the metabolic syndrome. This study was conducted to develop a rat model of metabolic syndrome by induction of fructose drinking water in different percentage concentrations of 20% and 25%. Activity and expression of 11β-HSD1 enzyme and metabolic syndrome complications such as NAFLD and pancreatic steatosis were also studied. Twenty percent and 25% fructose drinking water was given daily for 8 weeks as an ad libitum to male Wistar rats in addition to the normal rats' diet. Results of this study showed that consumption of 20% and 25% fructose drinking water for 8 weeks resulted in the formation of metabolic syndrome in male Wistar rats as well as increased the activity and expression of 11β-HSD1 enzyme. Complications of NAFLD and pancreatic steatosis can be seen in both groups. In conclusion, the lower fructose concentration which is 20% fructose drinking water which fulfills the criteria of metabolic syndrome as it gives significantly higher total daily fluid intake, total caloric intake, percentage of body weight gain, body mass index, abdominal circumference, abdominal adipose tissue depositions and weight than with that of 25% fructose drinking water was taken to establish a model of metabolic syndrome and complications of NAFLD and pancreatic steatosis in male Wistar rats through increased activity and expression of 11β-HSD1 enzyme mechanism.,Sarjana Sains Perubatan
