Description: | The extract of Uncaria gambir consists of catechin, which has been proven to exert antibacterial, anti-inflammatory and antioxidant effects. These desirable properties suggest that the extracts may have a role in managing dental caries and pulp therapy. However, in the literature, the study on the effect of Uncaria gambir extract against oral micro-flora is scarce and the methodology is not well documented. Therefore, we conducted this in vitro study to determine the antibacterial effects of Uncaria gambir extracts against four different bacteria; Streptococcus mutans, Streptococcus sobrinus, Enterococcus faecalis and Lactobacillus casei. The minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) value were determined using broth microdilution technique. Structural changes of Uncaria gambir-treated bacteria were visualised under transmission electron microscope (TEM). The inhibitory ability of the extracts to supress the bacterial growth were determined using time kill analysis. Both Streptococcus mutans and Streptococcus sobrinus showed MIC and MBC value at 1.25 mg/ml and 5 mg/ml respectively. In contrast, Enterococcus faecalis and Lactobacillus casei showed a higher MIC values of 2.5 mg/ml and 7.5 mg/ml and MBC value of 10 mg/ml and 30 mg/ml respectively. No statistically significant difference observed in the MIC values of Uncaria gambir extract against all bacteria as compared to 0.12% chlorhexidine gluconate mouthwash, with p>0.05. Destruction of the bacterial cell wall were noticed when viewed using TEM for all bacterial species. The Uncaria gambir extracts supressed the bacterial activity for all four bacterial species throughout the 24 hours of study period in a consistent manner. In conclusion, the Uncaria gambir extract exerts antibacterial effects against Streptococcus mutans, Streptococcus sobrinus, Enterococcus faecalis and Lactobacillus casei as shown through the MIC and MBC value, TEM and time kill analysis, with Streptococcus mutans and Streptococcus sobrinus were the most susceptible bacteria,Ijazah Doktor Pergigian Klinikal (Pergigian Pediatrik) |
