dc.description | Zingiber zerumbet has been used traditionally to treat various ailments include immunerelated diseases such as asthma, allergy, edema and arthritis in Malaysia and many other countries. It has been reported to possess various pharmacological properties including anti-inflammatory, anti-cancer, antioxidant, antiproliferative and anti-nociceptive activities. However, there is little scientific information regarding the immunological effects and immunomodulatory activity of Z. zerumbet in animal model. This study was carried out to investigate the immunomodulatory properties of standardized 80% ethanol extract of Z. zerumbet on innate and adaptive immune responses in Wistar rats. The extract was administered orally to male Wistar rats (100, 200 and 400 mg/kg of body weight) for 15 days for determination of innate immune responses, whereas for determination of adaptive responses, it was given for 21 days and immunized with 200 ?L of 5 x 109 sheep red blood cells (sRBCs) per mL on day 15. Delayed-type hypersensitivity (DTH) study was performed to determine the effect of extract on swelling of footpad. There was 24.27% of bioactive marker (zerumbone) was identified and quantified in the extract by using a validated reversed-phase high performance liquid chromatography (RP-HPLC) method. The extract significantly inhibited the four major steps of phagocytosis processes in innate immune system including the migration of neutrophils, expressions of CD11b/CD18 integrin, phagocytic activity and production of reactive oxygen species (ROS) in neutrophils. Lysozyme and ceruloplasmin expressions in rat plasma were also reduced dose-dependently. The proliferation of spleen lymphocytes, production of T subpopulations, Th1 and Th2 cytokines (IFN-γ, IL-2, IL-4) expression, and liberation of antibodies (IgM and IgG) in plasma were markedly suppressed in extract-treated rats. In DTH study, the extract significantly reduced footpad edema with more than 60% inhibition by the 200 and 400 mg/kg dose groups when compared to normal control group. Z. zerumbet extract possessed strong inhibitory effects on the immune responses and has potential to be developed into an effective immunosuppressive agent for the prevention and treatment of immune-related diseases.,The Degree of Master in Science (Pharmacy) | - |
